Product Information: The iLite™ IFN-Alpha Kit is based upon conventional gene-reporter assay format that has been modified and adapted for the quantitative determination of biologically active IFN-Alpha levels. Normal IFN-Alpha sensitive gene has been carefully selected and replaced with firefly luciferase with modifications to maximize reporter gene expression. Features
•Gene reporter assay format works with serum,
plasma, or culture supernatants
The reporter cells used in the kit have been selected for a high level of active IFN-Alpha receptor protein and are grown in a controlled manner that the number of receptor cell is constant. They are then treated by proprietary method to render them "cell proliferation deficient". These division arrested cells respond in a predictable manner when used according to the assay instructions. The iLite ™ IFN-Alpha Kit features an ease of use method for quantitative determination of IFN-Alpha level. To quantify the amount of IFN-Alpha in a biological sample, simply remove the kit from freezer, thaw the reporter cells and distribute to a 96-well plate, and add samples and IFN-Alpha standard. The cells are then incubated for 4-18 hours. At this time, the amount of active enzyme molecules is directly related to the number of IFN-Alpha molecules that have bound to the cell receptor. The luminescent substrates for the enzyme are then added and the reaction measured in a conventional luminometer. The level of luciferase enzyme activity observed in the assay can be correlated to International Units (IU) of the IFN-Alpha by generation of a standard curve using the supplied IFN-Alpha standard.
